Abstrakt

Předložená práce je zaměřena na detekci nitrifikačních bakterií ve vzorcích z českých a zahraničních čistíren odpadních vod metodou fluorescenční in situ hybridizace. Cílem práce bylo zjistit, jaké druhy nitrifikačních bakterií se na vybraných čistírnách odpadních vod vyskytují, zjistit základní kvantifikační parametry jednotlivých nitrifikačních populací v souvislosti s typem intenzifikace nitrifikace a ověřit, zda se tyto parametry významně liší od parametrů nitrifikačních populací čistíren bez podpory nitrifikace.

Teoretická část se zabývá problematikou vypouštění odpadních vod v souvislosti s legislativou. Popsána je obecná charakteristika jednotlivých nitrifikačních rodů a druhů zodpovědných za oxidaci amoniaku na dusitany a oxidaci dusitanů na dusičnany. Dále se teoretická část zabývá možností stanovení a identifikace nitrifikačních bakterií kultivačními technikami a metodami nezávislými na kultivaci, zejména metodami molekulárně-biologickými. Značná pozornost je věnována hybridizačním technikám a to jak jejich historii, tak citlivosti těchto metodik, výhodám a úskalím, které tyto techniky přinášejí, principu a postupu a dále pak možnosti kombinace s jinými technikami. Z metod intenzifikace nitrifikace na čistírnách odpadních vod je popsána bioaugmentace in situ. Teoretická část se zabývá rovněž využitím techniky fluorescenční in situ hybridizace pro detekci bakterií na čistírnách odpadních vod a to nejenom pro nitrifikační bakterie, ale rovněž pro bakterie zodpovědné za odstraňování fosforu a pro vláknité bakterie.

V experimentální části je popsán postup metodiky fluorescenční in situ hybridizace (včetně přípravy potřebných roztoků). Dále se experimentální část zabývá postupem vyhodnocení mikroskopického obrazu pomocí programů Lucia G a daime. V této části jsou popsány uspořádání a parametry jednotlivých sledovaných čistíren a způsob odběru vzorků.

Kapitola Výsledky a diskuze je věnována problematice vzorku aktivovaného kalu z hlediska zpracování metodikou fluorescenční in situ hybridizace a z hlediska interpretace získaných dat. Kapitola se zabývá rovněž morfologií a velikostí nitrifikačních klastrů ve zkoumaných vzorcích. Jsou zde shrnující tabulky a grafy s naměřenými daty pro jednotlivé čistírny odpadních vod včetně základních statistických veličin. Získané výsledky jsou pak diskutovány na základě intenzifikace nitrifikace, velikosti čistírny a stáří kalu.

Naměřená data ukazují, že hlavními bakteriemi zodpovědnými za první stupeň nitrifikace, tj. oxidace amoniaku na dusitany jsou bakterie oxidující amoniak patřící do skupiny ß-Proteobacteria a za druhý stupeň nitrifikace, tj. oxidace dusitanů na dusičnany bakterie rodu Nitrospira. U čistírny vybavené nitrifikačním a denitrifikačním biofiltrem byl zjištěn významný podíl bakterií rodu Nitrosomonas jako zástupci zmíněné skupiny amoniak-oxidujících ß-proteobakterií. Bakterie rodu Nitrosomonas byly detekované i v jedné z čistíren s regenerací (avšak v daleko menší míře), dále pak u jedné z aktivační zahraničních ČOV, (pouze v jednom vzorku). V aktivačních zahraničních čistírnách byl výjimečně, v malém množství, detekován rod Nitrosospira. Tyto bakterie jako jediné byly přítomny pouze ve formě samostatných buněk, ostatní detekované nitrifikační bakterie vytvářely rozmanité klastry různých tvarů a velikostí. Na základě získaných výsledků pro všechny sledované ČOV lze konstatovat, že na čistírnách s regenerací převládají bakterie rodu Nitrospira nad ß-Proteobacteria oxidujících amoniak. Na čistírně s biofiltrem a aktivačních ČOV bez podpory nitrifikace tomu bylo naopak. Nebyl prokázán vliv ročního období na druhové zastoupení nitrifikačních bakterií a na naměřené parametry nitrifikačních klastrů. Analýza mikroskopického obrazu odhalila velkou variabilitu naměřených veličin (povrch a objem klastrů, objemové zastoupení nitrifikační populace v celkové populaci bakterií) pro jednotlivá mikroskopická pole jednoho vzorku.

Abstract

This study is focused on the detection of nitrifying bacteria in Czech and foreign wastewater treatment plant samples by fluorescence in situ hybridization. The aim was to determine the type of nitrifiers and their basic parameters in selected wastewater treatment plants (WWTP) in context with the type of the intensification of nitrification and to check if population of nitrifying bacteria present on WWTP without the intensification of nitrifiacation is significantly different.

Theoretical part is oriented on legal questions of wastewater disposal. A general characterization of nitrifying genera and species responsible for the oxidation of ammonia to nitrite and the oxidation of nitrite to nitrate is described as well as both cultivation-dependent and independent methods (based on molecular biology) used for the identification of nitrifying bacteria. A considerable attention is paid to hybridization techniques, namely to their history, to the sensitivity of these methods, and to advantages and difficulties brought by these methods as well as to their combination with other techniques. In situ bioaugmentation method of intensification of nitrification right on wastewater treatment plants is described. A fluorescence in situ hybridization method used for the detection of not only nitrifying bacteria but also for the detection of bacteria responsible for the phosphorus removal and for the detection of filamentous bacteria on wastewater treatment plants is also part of this study.

In the experimental part a fluorescence in situ hybridization is described in detail, while the process of the evaluation of microscopic images supported by programs Lucia G and daime and the arrangement and parameters of monitored wastewater treatment plants and specific sampling procedures are also mentioned.

A chapter Results and Discussion is devoted to problems of activated sludge samples from the point of view of processing by fluorescence in situ hybridization and from the point of view of the interpretation of obtained data. This chapter also deals with the morphology and the size of nitrifying clusters in monitored samples. Summary tables and graphs with measured data for single wastewater treatment plants including basic statistical characteristics are presented. The obtained results are discussed based on the intensification of nitrification, the population equivalent and the sludge age.

Gained results show that bacteria responsible for the first step of nitrification, i. e. for the oxidation of ammonia to nitrite, are ammonia-oxidizing bacteria belonging into the genus of b-Proteobacteria and bacteria responsible for the second step of nitrification, i. e. for the oxidation of nitrite to nitrate belong to genus Nitrospira. Wastewater treatment plant with nitrifying and denitrifying filter was abundantly colonized with species Nitrosomonas sp. as representative of mentioned ammonia-oxidizing b-Proteo-bacteria. Bacteria of genus Nitrosomonas were also detected on one waste water treatment plant supplied with regeneration (but not in high levels) and in one sample from the wastewater treatment plant with the activation but without the intensification of nitrification. Nitrosospira sp. were also rarely detected in foreign wastewater treatment plants with activation. While bacteria Nitrospira sp. were presented in the form of single cells, the other ones created various clusters of different shapes and sizes. On the bases of gained results for all monitored wastewater treatment plants it is possible to note that on wastewater treatment plants with regeneration predominate bacteria of genus Nitrospira over ammonia-oxidizing b-Proteobacteria. The opposite tendency was observed on the wastewater treatment plant with the biofilter and on wastewater treatment plants with the activation but without the intensification of nitrification. The influence of season over generic nitrifiers composition and over measuring parameters of nitrifying clusters was not proved. The image analysis disclosed a great variability of measured quantities (area and volume of nitrifying clusters, volume fraction of nitrifying bacteria in the whole bacterial population) for single microscopic fields of one sample.
