Thesis topic:

Application of on-line biosensors in monitoring of drinking water quality

Motivation

The quality of drinking water is determined by the valid legislation which ensures its health safety for the consumer. Currently, some online monitoring system for microbial contamination is missing in the field of water treatment. Cultivation methods are mostly used for the monitoring of the microbial quality of drinking water. But their main disadvantage is the time to consume, more precisely the time needed for the incubation of cultivated microorganisms. If the microbial contamination of drinking water occurs, the identification of pollution in time is important, especially for pathogenic microorganisms which are the cause of a wide range of diseases. The aim is, therefore, to develop and implement the previously mentioned on-line measuring system for monitoring microbial contamination in real time.

Solution

The solution is the development and application of ultrasensitive electrochemical biosensors, where the basis is made from boron-doped nanocrystalline diamond layer ("BDNCD"), which is immobilized by sensors biomolecules such as antibodies, proteins, carbohydrates, or DNA oligonucleotides. These biomolecules are able to interact with bacterial cell surface molecules, which, together with the unique electrochemical properties of the BDNCD layer, provides the biosensors with a high detection sensitivity. Since Escherichia coli is an indicator of the fecal contamination of drinking water, it has also been chosen as a model organism for the development of this biosensor.
